How to read an insulin syringe
Pick your syringe, then drag the plunger or type a number. You’ll see the line it sits on, the mL, and what every small line on a 30, 50 or 100-unit U-100 syringe is worth.

How our calculator worksType a number of units
Add the mg/mL of your mixed vial to see how much this draw holds.
30, 50 and 100-unit syringes compared
All three are U-100 insulin syringes: 100 units fill 1 mL, whatever the size. They differ in how much they hold and how finely they’re marked.
| Syringe | Holds | Each line | Numbers |
|---|---|---|---|
| 30-unit | 0.3 mL | 1 unit | every 5 |
| 50-unit | 0.5 mL | 1 unit | every 5 |
| 100-unit | 1 mL | 2 units | every 10 |
Needles differ too, in length and gauge. See insulin syringe and needle sizes.
What each tick mark means
Count the small lines between two numbers. On 30 and 50-unit syringes there are 5 steps between numbers, so each line is 1 unit (0.01 mL). On a 100-unit syringe there are 5 steps between numbers 10 apart, so each line is 2 units (0.02 mL).
Half-unit markings
Some 30-unit syringes add half-unit lines; most syringes don’t. Always read the marks on your own syringe. When a calculated draw falls between lines, our calculators show the nearest line and how much it changes the amount.
Reading the plunger edge
Line up the top edge of the black rubber plunger tip, the flat ring nearest the needle, with your number (MedlinePlus). Hold the syringe at eye level and tap out air bubbles before you check.
Need the units for a dose first? The peptide calculator works them out from your vial, water and prescribed dose.
Questions people ask
What does U-100 mean on an insulin syringe?
U-100 means the scale is made for liquids measured at 100 units per mL. Every mark is a volume: 1 unit is 0.01 mL, so 100 units is 1 mL, 50 units is 0.5 mL and 30 units is 0.3 mL. Peptinote’s calculators all use U-100 syringes; U-40 and U-500 syringes use different scales.
What’s the difference between 30, 50 and 100-unit syringes?
How much they hold and how finely they’re marked. A 30-unit syringe holds 0.3 mL and a 50-unit syringe 0.5 mL, both with a line for every unit and numbers every 5. A 100-unit syringe holds 1 mL, with a line every 2 units and numbers every 10, so small or odd amounts are harder to read on it.
How do I read half units on an insulin syringe?
Only some syringes have half-unit lines, usually 30-unit ones; check the barrel of yours. On a syringe without them, a half unit sits between two lines and can’t be measured exactly. Our calculators show the nearest printed line and how much that changes the amount, so you can ask your pharmacist how to handle it.
Where on the plunger do I read the units?
At the top edge of the black rubber plunger tip, the flat ring nearest the needle. Line that edge up with your number, not the pointed tip or the bottom of the rubber. MedlinePlus describes the same method. Tap out air bubbles first: they push the plunger past your line.
Can Peptinote tell me how many units to take?
No. Peptinote never suggests a dose. Enter the dose your prescriber gave you, and the calculator shows how to measure it: the units, the mL and where the line sits on your syringe, with every step of the math. If you aren’t sure what your dose is, ask your prescriber or pharmacist before drawing anything. How our calculator works.
